Transcriptional regulation at the single cell level

I am interested in the regulation of gene expression in the yeast Saccharomyces cerevisiae. When yeast sense stress in the environment they initiate a broad gene expression response and I am interested in how this is regulated. Although many genes are under the transcriptional control of a single transcription factor, it is not entirely clear how it is possible for a single transcription factor to differentially regulate the expression of hundreds of different genes. Specifically, I am interested in how the stress response initiated by Msn2 is decoded by the genes that show elevated expression in an Msn2-dependent fashion. Previous work in our lab has generated tools to control the activity and dynamics of Msn2. I hope use this tool and to map the promoter structure of Msn2-responding genes and us this to elucidate how this allows for differential gene expression in response to stress.
