Colony PCR


  • Streak out yeast and grow until small colonies form.


  1. Place 5 ul of water into a PCR tube.
  2. Stick a pipette tip into the colony to pick up a small number of cells. Smash the pipette tip into the cells.
  3. Freeze the PCR tube for 10-15 min.
  4. Microwave the PCR tube for 1 min on high power.
  5. Freeze the PCR tube for 10-15 min.
  6. Prepare 45 ul of the PCR mix.
  7. Add the PCR mix to the tube, mix, spin down, and PCR as normal.
